Determination of specific IgE to two major allergens(Der f1 and Der f2) of house dust mite(D. farinae) in Korean adult respiratory allergy patients



Abstract
House dust mite has been known as a most important allergen in allergic respiratory diseases. More than 30 allergenic components were confirmed in the whole body extract of D. farinae. But among these allergens, Der fI and Der fII were recognized
as
major allergens. In this study, we measured the specific IgE to WBE. Der fI and fII on ELISA in 134 adults with history of allergic respiratory diseases and the results were as follwed.
1. In the 18 cases of control group, mean O. D. value of specific IgE to WBE, Der fI and Der fII were 0.011¡¾0.004/0.004¡¾0.002/0.007¡¾0.002 respectively. So the O.D. value 0.020 was chosen as the cut-off value of Der fI and Der fII.
2. Among the 31 cases with negative skin responsiveness to D. farinae allergen, specific IgE to WBE was detected in 5 cases(16%) and specific IgE to Der fI and fII was detected in 1 case each. But in 103 cases with positive skin responsiveness
to
D.
farinae allergen, specific IgE to EBE, Der fI and Der fII were found in 86(83%), 62 (60%) and 63(61%) cases respectively. Further more we could find significant relationship between the skin responsiveness and the detection rate of specific IgE
to
WBE,
Der fII in 134 respiratory allergy patients.
3. The O.D. value of specific IgE to WBE/Der fI /Der fII were 0.014/0.005/0.05 in 31 cases of negative group to skin prick test of D.farinae, 0.022/0.009/0.013 for 1+ group, 0.069/0.009/0.013 for 2+ group, 0.212.0.013.0.172 for 3+ group,
0.0780.0.0110/0.687 for 4+ group, and 0.712/0.150/0.845 for 5+ group. Therefore, the O.D. value of specific IgE to Der fII was much higher than that of Der fI in strong skin reactive groups.
4. With the 20§¶ of WBE, the specific IgE to WBE, Der fI and Der fII were inhibited. In case of inhibition by 20ng of Der fI, the specific IgE to Der fI was specifically inhibited but the specific IgE to WBE and Der fII were not inhibited. With
the
inhibition by 20ng of Der fII, we could find significant inhibiton of specific IgE to Der fII and WBE but the specific IgE to Der fI was not inhibited.
In conclusion, both of Der fI and Der fII were recognized as important major allergenic components of D. farinae in Korean adult allergic respiratory diseases. And we could suggest that the role of Der fII was more important in pathophysiology of
allergic respiratory diseases.
